Functional DNA Nanostructures-Gold Nanoparticles
for Targeted Gene Therapy Purposes

General Objective
The main objective of the proposal is to design and prepare nano platform for the targeted gene therapy using functionalized gold nanoparticles positioned on a DNA nanostructure.
Specific Objectives

To design and synthesize gold nanoparticles decorated with cationic polymer and PEG-ylated cyclodextrin molecules.

To establish methodology for the quantification of the amount of PEI/PEG-Cy moieties on the surface of AuNPs.

To position functionalized AuNPs on DNA origami nanostructure. Free cyclodextrin molecules on the AuNPs surface and multiple grouped adamantane molecules on the surface of DNA nanostructure will be utilized.

To perform cytotoxicity and in vitro studies to test the transfection efficiency in four available cancer cells.
